🧫#CellCulture101: Mastering Cell Dissociation Proper cell dissociation is key to successful cell culture. Timing is crucial; overly digesting cells can cause deterioration while under-digesting can hinder your experiments. When to dissociate cells: ◾ Digest cells at 80%-90% confluence. ◾ Use digestion reagents to detach cells from the culture vessel for passage. 𝐌𝐞𝐭𝐡𝐨𝐝𝐬 𝐨𝐟 𝐂𝐞𝐥𝐥 𝐃𝐢𝐬𝐬𝐨𝐜𝐢𝐚𝐭𝐢𝐨𝐧: ◽ Enzymatic: Trypsin is commonly used, with digestion time depending on cell type and temperature. ◽ Chelating agents: EDTA chelates cell adhesion molecules, useful when analyzing cell surface molecules. ◽ Physical: Tap or scrape cells off using a cell scraper. ◽ Freezing: Minimal damage to cells, but more suited for cell passaging. 𝐒𝐢𝐠𝐧𝐬 𝐨𝐟 𝐚 𝐬𝐮𝐜𝐜𝐞𝐬𝐬𝐟𝐮𝐥 𝐝𝐢𝐬𝐬𝐨𝐜𝐢𝐚𝐭𝐢𝐨𝐧: ◾ Cells should move freely under the microscope, indicating adhesion loss. ◾ Not all cells will become round; some may appear semi-sandy. ◾ Watch out for detached cells; this may suggest incomplete digestion. 𝐇𝐨𝐰 𝐭𝐨 𝐡𝐚𝐧𝐝𝐥𝐞 𝐨𝐯𝐞𝐫-𝐝𝐢𝐠𝐞𝐬𝐭𝐢𝐨𝐧: ◽ Neutralize with culture medium and centrifuge cells at 800 RPM for 3 minutes. ◽ Resuspend in fresh medium and continue culturing.
